The CVI Antibody Core Providing expertise and access to the ForteBio OctetÒ RED384 equipment to the Athens and Atlanta Research community.

The Octet system provides researchers the opportunity to perform high - throughput, real - time , drug - protein and protein - protein binding kinetics in an expedited manner compared to traditional techniques.

The high - throughput aspect of the system means that a library of small molecules or known proteins can be screened for interaction with a protein of interest both quickly and at large scale. The BioLayer Interferometry (BLI) Technology provides real - time protein - small molecule or protein - protein association and dissociation kin etics. T he high sensitivit y of the assay means that much lower protein amounts are required for analysis compared to traditional immunoprecipitation , NMR, or ELISA methods. In addition, crude pr otein extracts may be used, and the OctetÒ system has considerable flexibility to work with both labeled and non - labeled protein targets.

ForteBio’s Octet® platform includes instruments, biosensors, reagents and assay kits for performing label-free, real-time protein quantitation and biomolecular binding kinetic analysis. in 96- or 384-well microplates.

    • Biomolecular Kinetics: The Octet accurately measures kinetic constants by bringing the detection surface directly to the sample, eliminating the need for micro-fluidics. This approach utilizing label-free, real-time analysis allows direct measurement of crude samples.
    • Affinity Measurements — ka, kd, KD : Real-time kinetic measurements offer a direct realistic depiction of molecular interactions. The Octet-384 is label-free system for full affinity determination, enabling users to easily and accurately obtain kinetic constants such as ka, kd, KD.
    • Kinetic Screening and Clone Selection: Researchers looking to select the best clones from a large pool will reap substantial benefits with the Octet due to its throughput and crude sample compatibility.
    • Epitope Binning: When multiple candidates have similar affinities for a target, determining their preference for the same or different epitopes on the target molecule becomes a crucial factor in their evaluation. Separating the candidates into groups (bins) with a common preference for the same epitope helps researchers further characterize the specificity of the clones and their ability to block target activity.
    • Ligand Capture: With a wide range of ligand capture surfaces that allow for the immobilization of antibodies, recombinant proteins, DNA, RNA and virus like particles (VLPs), ForteBio's Octet 384 provides real-time measurements in the analysis of therapeutic biological molecules at different stages of development. The applications include but are not limited to protein:protein, protein:nucleic acids, protein:small molecules interactions and the analysis of large molecules such as virus-like particles.
